Abstract: The accumulation of anthocyanins in plants exposed to salt stress has been largely documented. However, the functional link and regulatory components underlying the biosynthesis of these molecules during exposure to stress are largely unknown. In a screen of second site suppressors of the salt overly sensitive3-1 (sos3-1) mutant, we isolated the anthocyanin-impaired-response-1 (air1) mutant. air1 is unable to accumulate anthocyanins under salt stress, a key phenotype of sos3-1 under high NaCl levels (120 mM). The air1 mutant showed a defect in anthocyanin production in response to salt stress but not to other stresses such as high light, low phosphorous, high temperature or drought stress. This specificity indicated that air1 mutation did not affect anthocyanin biosynthesis but rather its regulation in response to salt stress. Analysis of this mutant revealed a T-DNA insertion at the first exon of an Arabidopsis thaliana gene encoding for a basic region-leucine zipper transcription factor. air1 mutants displayed higher survival rates compared to wild-type in oxidative stress conditions, and presented an altered expression of anthocyanin biosynthetic genes such as F3H, F3’H and LDOX in salt stress conditions. The results presented here indicate that AIR1 is involved in the regulation of various steps of the flavonoid and anthocyanin accumulation pathways and is itself regulated by the salt-stress response signalling machinery. The discovery and characterization of AIR1 opens avenues to dissect the connections between abiotic stress and accumulation of antioxidants in the form of flavonoids and anthocyanins.

Abstract: The human population is projected to reach more than 10 billion in the year 2100. Together with changing consumption pattern, population growth will lead to increasing food demand. The question arises whether or not the Earth is capable of fulfilling this demand. In this study, we approach this question by estimating the carrying capacity of current agricultural systems (K-C), which does not measure the maximum number of people the Earth is likely to feed in the future, but rather allows for an indirect assessment of the increases in agricultural productivity required to meet demands. We project agricultural food production under progressing climate change using the state-of-the-art dynamic global vegetation model LPJmL, and input data of 3 climate models. For 1990 to 2100 the worldwide annual caloric yield of the most important 11 crop types is simulated. Model runs with and without elevated atmospheric CO2 concentrations are performed in order to investigate CO2 fertilization effects. Country-specific per-capita caloric demands fixed at current levels and changing demands based on future GDP projections are considered to assess the role of future dietary shifts. Our results indicate that current population projections may considerably exceed the maximum number of people that can be fed globally if climate change is not accompanied by significant changes in land use, agricultural efficiencies and/or consumption pathways. We estimate the gap between projected population size and K-C to reach 2 to 6.8 billion people by 2100. We also present possible caloric self-supply changes between 2000 and 2100 for all countries included in this study. The results show that predominantly developing countries in tropical and subtropical regions will experience vast decreases of self-supply. Therefore, this study is important for planning future large-scale agricultural management, as well as the critical assessment of population projections, which should take food-mediated climate change feedbacks into account

Abstract: Ectopic cystatin expression has long been used in plant pest management, but the cysteine protease, targets of these inhibitors, might also have important functions in the control of plant lifespan and stress tolerance that remain poorly characterized. We therefore characterized the effects of expression of the rice cystatin, oryzacystatin-I (OCI), on the growth, development and stress tolerance of crop (soybean) and model (Arabidopsis thaliana) plants. Ectopic OCI expression in soybean enhanced shoot branching and leaf chlorophyll accumulation at later stages of vegetative development and enhanced seed protein contents and decreased the abundance of mRNAs encoding strigolactone synthesis enzymes. The OCI-expressing A. thaliana showed a slow-growth phenotype, with increased leaf numbers and enhanced shoot branching at flowering. The OCI-dependent inhibition of cysteine proteases enhanced drought tolerance in soybean and A. thaliana, photosynthetic CO2 assimilation being much less sensitive to drought-induced inhibition in the OCI-expressing soybean lines. Ectopic OCI expression or treatment with the cysteine protease inhibitor E64 increased lateral root densities in A. thaliana. E64 treatment also increased lateral root densities in the max2-1 mutants that are defective in strigolactone signalling, but not in the max3-9 mutants that are defective in strigolactone synthesis. Taken together, these data provide evidence that OCI-inhibited cysteine proteases participate in the control of growth and stress tolerance through effects on strigolactones. We conclude that cysteine proteases are important targets for manipulation of plant growth, development and stress tolerance, and also seed quality traits.

Abstract: Given the energetic, demographic and the climatic challenges faced today, we designed a combined food and energy (CFE) production system integrating food, fodder and mixed belts of Salix, Alnus and Corylus sp. as bioenergy belts. The objective was to assess the shoot dry weight-stem diameter allometric relationship based on stem diameter at 10 (SD10) and 55 cm (SD55) from the shoot base in the mixed bioenergy belts. Allometric relations based on SD10 and SD55 explained 90-96% and 90-98% of the variation in shoot dry weights respectively with no differences between the destructive and the non-destructive methods. The individual stool yields varied widely among the species and within willow species with biomass yield range of 37.60-92.00 oven dry tons (ODT) ha (1) in 4-year growth cycle. The biomass yield of the bioenergy belt, predicted by allometric relations was 48.84 ODT ha 1 in 4-year growth cycle corresponding to 12.21 ODT ha (1) year (1). The relatively high biomass yield is attributed to the border effects and the ‘fertilizing effect’ of alder due to nitrogen fixation, benefitting other SWRC components. On termination of 4-year growth cycle, the bioenergy belts were harvested and the biomass yield recorded was 12.54 ODT ha (1) year (1), in close proximity to the biomass yield predicted by the allometric equations, lending confidence and robustness of the model for biomass yield determination in such integrated agro-ecosystem. Abstract: Intensified climate and market turbulence requires resilience to a multitude of changes. Diversity reduces the sensitivity to disturbance and fosters the capacity to adapt to various future scenarios. What really matters is diversity of responses. Despite appeals to manage resilience, conceptual developments have not yet yielded a break-through in empirical applications. Here, we present an approach to empirically reveal the ‘response diversity’: the factors of change that are critical to a system are identified, and the response diversity is determined based on the documented component responses to these factors. We illustrate this approach and its added value using an example of securing food supply in the face of climate variability and change. This example demonstrates that quantifying response diversity allows for a new perspective: despite continued increase in cultivar diversity of barley, the diversity in responses to weather declined during the last decade in the regions where most of the barley is grown in Finland. This was due to greater homogeneity in responses among new cultivars than among older ones. Such a decline in the response diversity indicates increased vulnerability and reduced resilience. The assessment serves adaptive management in the face of both ecological and socioeconomic drivers. Supplier diversity in the food retail industry in order to secure affordable food in spite of global price volatility could represent another application. The approach is, indeed, applicable to any system for which it is possible to adopt empirical information regarding the response by its components to the critical factors of variability and change. Targeting diversification in response to critical change brings efficiency into diversity. We propose the generic procedure that is demonstrated in this study as a means to efficiently enhance resilience at multiple levels of agrifood systems and beyond. (C) 2014 The Authors.
