Abstract: The accumulation of anthocyanins in plants exposed to salt stress has been largely documented. However, the functional link and regulatory components underlying the biosynthesis of these molecules during exposure to stress are largely unknown. In a screen of second site suppressors of the salt overly sensitive3-1 (sos3-1) mutant, we isolated the anthocyanin-impaired-response-1 (air1) mutant. air1 is unable to accumulate anthocyanins under salt stress, a key phenotype of sos3-1 under high NaCl levels (120 mM). The air1 mutant showed a defect in anthocyanin production in response to salt stress but not to other stresses such as high light, low phosphorous, high temperature or drought stress. This specificity indicated that air1 mutation did not affect anthocyanin biosynthesis but rather its regulation in response to salt stress. Analysis of this mutant revealed a T-DNA insertion at the first exon of an Arabidopsis thaliana gene encoding for a basic region-leucine zipper transcription factor. air1 mutants displayed higher survival rates compared to wild-type in oxidative stress conditions, and presented an altered expression of anthocyanin biosynthetic genes such as F3H, F3’H and LDOX in salt stress conditions. The results presented here indicate that AIR1 is involved in the regulation of various steps of the flavonoid and anthocyanin accumulation pathways and is itself regulated by the salt-stress response signalling machinery. The discovery and characterization of AIR1 opens avenues to dissect the connections between abiotic stress and accumulation of antioxidants in the form of flavonoids and anthocyanins.

Abstract: Ultraviolet (UV) radiation, particularly in the UV-A + B range (280-400 nm) is a fraction of the solar spectrum that regulates almost every aspect of insect behaviour, including orientation towards hosts, alighting, arrestment and feeding behaviour. To study the role of UV radiation on the flight activity of five insect species of agricultural importance (pests Myzus persicae, Bemisia tabaci and Tuta absoluta, and natural enemies Aphidius colemani and Sphaerophoria rueppellii), one-chamber tunnels were covered with six cladding materials with different light transmittance properties ranging from 2% to 83% UV and 54% to 85% photosynthetically active radiation (PAR). Inside each tunnel, insects were released from tubes placed in a platform suspended from the ceiling. Specific targets varying with insect species were placed at different distances from the platform. Evaluation parameters were designed for each insect and tested separately. The ability of insects to leave the platform was assessed, as well as the number of captures, eggs or mummies in each target, either sticky traps or plants. Our results suggest differences in flight activity among insect species and UV-blocking nets. The UV-opaque film drastically prevented aphids, and whiteflies from flying outside the tubes whereas T. absoluta, syrphids and parasitoids were not affected. Aphid flight behaviour was affected by the UV-opaque film compared to the other nets, especially in the furthest target of the tunnel. Fewer aphids reached distant traps under UV-absorbing nets, and significantly more aphids could fly to the end of tunnels covered with non-UV-blocking materials. Orientation of B. tabaci and T. absoluta was also negatively affected by the UV-opaque film although in a different trend. Unlike aphids, differences in B. tabaci captures were mainly found in the closest targets. UV transmittance did not have any effects on parasitoids, and S. rueppellii, implying cues other than visual for these insects under our experimental conditions. Further effects of photoselective enclosures on greenhouse pests and their natural enemies are discussed.

Abstract: Process-based grassland models (PBMs) simulate growth and development of vegetation over time. The models tend to have a large number of parameters that represent properties of the plants. To simulate different cultivars of the same species, different parameter values are required. Parameter differences may be interpreted as genetic variation for plant traits. Despite this natural connection between PBMs and plant genetics, there are only few examples of successful use of PBMs in plant breeding. Here we present a new procedure by which PBMs can help design ideotypes, i.e. virtual cultivars that optimally combine properties of existing cultivars. Ideotypes constitute selection targets for breeding. The procedure consists of four steps: (1) Bayesian calibration of model parameters using data from cultivar trials, (2) Estimating genetic variation for parameters from the combination of cultivar-specific calibrated parameter distributions, (3) Identifying parameter combinations that meet breeding objectives, (4) Translating model results to practice, i.e. interpreting parameters in terms of practical selection criteria. We show an application of the procedure to timothy (Phleum pratense L.) as grown in different regions of Norway.

Abstract: Phytophthora infestans is one of the most destructive potato pathogens. Many factors influence the population structure of P. infestans, including migration, climate and type of potato cultivation. Here, we analyse 365 P. infestans isolates collected from three regions of Poland over three years. We determined mating type, mitochondrial haplotype, resistance to metalaxyl, virulence and polymorphism at 14 simple sequence repeat (SSR) loci. Analysis of SSR markers showed high genetic diversity associated with this population. Model-based structure analysis grouped 299 unique genotypes into four main clusters. The P. infestans isolates collected from the Mlochow region, which has the most intensive level of potato cultivation, formed a distinct cluster, indicating a strong effect of the cultivation system on pathogen population structure. Three clusters contained isolates with frequent presence of three alleles at one locus, which may affect their capacity for sexual reproduction and preserve groups of fit genotypes that propagate asexually. Abstract: The main aim of the paper is to show how, and how many, simulations carried out using the Land Surface Model UTOPIA (University of TOrino model of land Process Interaction with Atmosphere) are representative of the micro-meteorological conditions and exchange processes at the atmosphere/biosphere interface, with a particular focus on heat and hydrologic transfers, over an area of the Piemonte (Piedmont) region, NW Italy, which is characterized by the presence of many vineyards. Another equally important aim is to understand how much the quality of the simulation outputs was influenced by the input data, whose measurements are often unavailable for long periods over country areas at an hourly basis. Three types of forcing data were used: observations from an experimental campaign carried out during the 2008, 2009, and 2010 vegetative seasons in three vineyards, and values extracted from the freely available Global Land Data Assimilation System (GLDAS, versions 2.0 and 2.1). Since GLDAS also contains the outputs of the simulations performed using the Land Surface Model NOAH, an additional intercomparison between the two models, UTOPIA and NOAH, both driven by the same GLDAS datasets, was performed. The intercomparisons were performed on the following micro-meteorological variables: net radiation, sensible and latent turbulent heat fluxes, and temperature and humidity of soil. The results of this study indicate that the methodology of employing land surface models driven by a gridded database to evaluate variables of micro-meteorological and agronomic interest in the absence of observations is suitable and gives satisfactory results, with uncertainties comparable to measurement errors, thus, allowing us to also evaluate some time trends. The comparison between GLDAS2.0 and GLDAS2.1 indicates that the latter generally produces simulation outputs more similar to the observations than the former, using both UTOPIA and NOAH models.
