Abstract: Successive single day transfers of pot-grown wheat to high temperature (35/30°C day/night) replicated controlled environments, from the second node detectable to the milky-ripe growth stages, provides the strongest available evidence that the fertility of wheat can be highly vulnerable to heat stress during two discrete peak periods of susceptibility: early booting [decimal growth stage (GS) 41-45] and early anthesis (GS 61-65). A double Gaussian fitted simultaneously to grain number and weight data from two contrasting elite lines (Renesansa, listed in Serbia, Ppd-D1a, Rht8; Savannah, listed in UK, Ppd-D1b, Rht-D1b) identified peak periods of main stem susceptibility centered on 3 (s.e. = 0.82) and 18 (s.e. = 0.55) days (mean daily temperature = 14.3°C) pre-GS 65 for both cultivars. Severity of effect depended on genotype, growth stage and their interaction: grain set relative to that achieved at 20/15°C dropped below 80% for Savannah at booting and Renesansa at anthesis. Savannah was relatively tolerant to heat stress at anthesis. A further experiment including 62 lines of the mapping, doubled-haploid progeny of Renesansa × Savannah found tolerance at anthesis to be associated with Ppd-D1b, Rht-D1b, and a QTL from Renesansa on chromosome 2A. None of the relevant markers were associated with tolerance during booting. Rht8 was never associated with heat stress tolerance, a lack of effect confirmed in a further experiment where Rht8 was included in a comparison of near isogenic lines in a cv. Paragon background. Some compensatory increases in mean grain weight were observed, but only when stress was applied during booting and only where Ppd-D1a was absent.

Abstract: Decisions for agricultural management are taken at farm scale. However, such decisions may well impact upon regional sustainability. Two of the likely agricultural management responses to future challenges are extended use of irrigation and increased production of energy crops. The drivers for these are high commodity prices and subsidy policies for renewable energy. However, the impacts of these responses upon regional sustainability are unknown. Thus, we conducted integrated impact assessments for agricultural intensification scenarios in the federal state of Brandenburg, Germany, for 2025. One Irrigation scenario and one Energy scenario were contrasted with the Business As Usual (BAU) scenario. We applied nine indicators to analyze the economic, social and environmental effects at the regional, in this case district scale, which is the smallest administrative unit in Brandenburg. Assessment results were discussed in a stakeholder workshop involving 16 experts from the state government. The simulated area shares of silage maize for fodder and energy were 29%, 37% and 49% for the BAU, Irrigation, and Energy scenarios, respectively. The Energy scenario increased bio-electricity production to 41% of the demand of Brandenburg, and it resulted in CO2 savings of up to 3.5 million tons. However, it resulted in loss of biodiversity, loss of landscape scenery, increased soil erosion risk, and increased area demand for water protection requirements. The Irrigation scenario led to yield increases of 7% (rapeseed), 18% (wheat, sugar beet), and 40% (maize) compared to the BAU scenario. It also reduced the year-to-year yield variability. Water demand for irrigation was found to be in conflict with other water uses for two of the 14 districts. Spatial differentiation of scenario impacts showed that districts with medium to low yield potentials were more affected by negative impacts than districts with high yield potentials. In this first comprehensive sustainability impact assessment of agricultural intensification scenarios at regional level, we showed that a considerable potential for agricultural intensification exists. The intensification is accompanied by adverse environmental and socio-economic impacts. The novelty lies in the multiscale integration of comprehensive, agricultural management simulations with regional level impact assessment, which was achieved with the adequate use of indicators. It provided relevant evidence for policy decision making. Stakeholders appreciated the integrative approach of the assessment, which substantiated ongoing discussions among the government bodies. The assessment approach and the Brandenburg case study may stay exemplary for other regions in the world where similar economic and policy driving forces are likely to lead to agricultural intensification. Abstract: Ectopic cystatin expression has long been used in plant pest management, but the cysteine protease, targets of these inhibitors, might also have important functions in the control of plant lifespan and stress tolerance that remain poorly characterized. We therefore characterized the effects of expression of the rice cystatin, oryzacystatin-I (OCI), on the growth, development and stress tolerance of crop (soybean) and model (Arabidopsis thaliana) plants. Ectopic OCI expression in soybean enhanced shoot branching and leaf chlorophyll accumulation at later stages of vegetative development and enhanced seed protein contents and decreased the abundance of mRNAs encoding strigolactone synthesis enzymes. The OCI-expressing A. thaliana showed a slow-growth phenotype, with increased leaf numbers and enhanced shoot branching at flowering. The OCI-dependent inhibition of cysteine proteases enhanced drought tolerance in soybean and A. thaliana, photosynthetic CO2 assimilation being much less sensitive to drought-induced inhibition in the OCI-expressing soybean lines. Ectopic OCI expression or treatment with the cysteine protease inhibitor E64 increased lateral root densities in A. thaliana. E64 treatment also increased lateral root densities in the max2-1 mutants that are defective in strigolactone signalling, but not in the max3-9 mutants that are defective in strigolactone synthesis. Taken together, these data provide evidence that OCI-inhibited cysteine proteases participate in the control of growth and stress tolerance through effects on strigolactones. We conclude that cysteine proteases are important targets for manipulation of plant growth, development and stress tolerance, and also seed quality traits.
