Abstract: Phytophthora infestans is one of the most destructive potato pathogens. Many factors influence the population structure of P. infestans, including migration, climate and type of potato cultivation. Here, we analyse 365 P. infestans isolates collected from three regions of Poland over three years. We determined mating type, mitochondrial haplotype, resistance to metalaxyl, virulence and polymorphism at 14 simple sequence repeat (SSR) loci. Analysis of SSR markers showed high genetic diversity associated with this population. Model-based structure analysis grouped 299 unique genotypes into four main clusters. The P. infestans isolates collected from the Mlochow region, which has the most intensive level of potato cultivation, formed a distinct cluster, indicating a strong effect of the cultivation system on pathogen population structure. Three clusters contained isolates with frequent presence of three alleles at one locus, which may affect their capacity for sexual reproduction and preserve groups of fit genotypes that propagate asexually. Abstract: The study was based on data collected during 5 yr (2003-2007) and was aimed at assessing the effects of the month, slaughter house of destination (differing for stocking density, openings, brightness, and cooling device types), length of the journey, and temperature-humidity index (THI) on mortality of heavy slaughter pigs (approximately 160 kg live weight) during transport and lairage. Data were obtained from 24,098 journeys and 3,676,153 pigs transported from 1,618 farms to 3 slaughter houses. Individual shipments were the unit of observation. The terms dead on arrival (DOA) and dead in pen (DIP) refer to pigs that died during transport and in lairage at the abattoir before slaughtering, respectively. These 2 variables were assessed as the dependent counts in separate univariate Poisson regressions. The independent variables assessed univariately in each set of regressions were month of shipment, slaughter house of destination, time traveled, and each combination of the month with the time traveled. Two separate piecewise regressions were done. One used DOA counts within THI levels over pigs transported as a dependent ratio and the second used DIP counts within THI levels over pigs from a transport kept in lairage as a dependent ratio. The THI was the sole independent variable in each case. The month with the greatest frequency of deaths was July with a risk ratio of 1.22 (confidence interval: 1.06-1.36; P < 0.05) and 1.27 (confidence interval: 1.06-1.51; P < 0.05) for DOA and DIP, respectively. The lower mortality risk ratios for DOA and DIP were recorded for January and March (P < 0.05). The aggregated data of the summer (June, July, and August) versus non-summer (January, March, September, and November) months showed a greater risk of pigs dying during the hot season when considering both transport and lairage (P < 0.05). The mortality risk ratio of DIP was lower at the slaughter house with the lowest stocking density (0.64 m(2)/100 kg live weight), large open windows on the roof and sidewalls, low brightness (40 lx) lights, and high-pressure sprinklers as cooling devices. The mortality risk ratio of DOA increased significantly for journeys longer than 2 h, whereas no relationship was found between length of transport and DIP. The piecewise analysis pointed out that 78.5 and 73.6 THI were the thresholds above which the mortality rate increased significantly for DOA and DIP, respectively. These results may help the pig industry to improve the welfare of heavy slaughter pigs during transport and lairage.

Abstract: The accumulation of anthocyanins in plants exposed to salt stress has been largely documented. However, the functional link and regulatory components underlying the biosynthesis of these molecules during exposure to stress are largely unknown. In a screen of second site suppressors of the salt overly sensitive3-1 (sos3-1) mutant, we isolated the anthocyanin-impaired-response-1 (air1) mutant. air1 is unable to accumulate anthocyanins under salt stress, a key phenotype of sos3-1 under high NaCl levels (120 mM). The air1 mutant showed a defect in anthocyanin production in response to salt stress but not to other stresses such as high light, low phosphorous, high temperature or drought stress. This specificity indicated that air1 mutation did not affect anthocyanin biosynthesis but rather its regulation in response to salt stress. Analysis of this mutant revealed a T-DNA insertion at the first exon of an Arabidopsis thaliana gene encoding for a basic region-leucine zipper transcription factor. air1 mutants displayed higher survival rates compared to wild-type in oxidative stress conditions, and presented an altered expression of anthocyanin biosynthetic genes such as F3H, F3’H and LDOX in salt stress conditions. The results presented here indicate that AIR1 is involved in the regulation of various steps of the flavonoid and anthocyanin accumulation pathways and is itself regulated by the salt-stress response signalling machinery. The discovery and characterization of AIR1 opens avenues to dissect the connections between abiotic stress and accumulation of antioxidants in the form of flavonoids and anthocyanins.

Abstract: Farmlands are increasingly exposed to degradation phenomena associated to climate change and agricultural practices, including irrigation. It is estimated that about 20% of the world’s irrigated land is salt affected. In this paper we aimed at evaluating the effect of seasonal and multiannual soil satinization on growth, yield, and radiation use efficiency of tomato in open field. Two field experiments were carried out at the Experimental Station of the University of Naples Federico II (latitude 40 degrees 31’N longitude 14 degrees 58’E) (Italy) on tomato during 2004 and 2005 to study the effect of five levels of water salinity: NSC (EC = 0.5 dS m(-1)), SW1 (EC= 2.3 dS m(-1)), SW2 (EC= 4.4 dS m(-1)), SW3 (EC= 8.5 dS m(-1)) and SW4 (EC= 15.7 dS m(-1)) in a soil exposed to one-season salinization (ST = short-term) and an adjacent soil exposed to >20 years salinization (LT = long-term). Plant growth, yield and fruit quality (pH, EC, total soluble solids and the concentration of reducing sugars and of titratable acids), and plant water relations were measured and radiation use efficiency (RUE) was calculated. Increasing water salinity negatively affected the leaf area index (LAI), radiation use efficiency (RUE) and above-ground dry weight (DW) accumulation resulting in lower total and marketable yield. Maximum total and marketable yield obtained with the NSC treatment were respectively 117.9 and 111.0 Mg ha(-1) in 2004 and 113.1 and 107.9 Mg ha(-1) in 2005. Although the smaller leaf area of salinized plants was largely responsible for reduced RUE, we found approximately 50% of this reduction to be accounted for by processes other than changed crop architecture. These may include an increased stomatal resistance, increased mesophyll resistance and other impaired metabolic functions that may occur at high salinity. Remarkably, we found that LT salinized plants had a slightly better efficiency of use of intercepted radiation (RUEIR) at a given EC of soil extract than ST salinized plants indicating that LT salinization, and consequent permanent modifications of the soil physical properties, may trigger additional physiological mechanisms of adaptation compared to ST salinized plants. These differences are relevant in light of the evolution of salinized areas, also in response to climate change.

Abstract: Ultraviolet (UV) radiation, particularly in the UV-A + B range (280-400 nm) is a fraction of the solar spectrum that regulates almost every aspect of insect behaviour, including orientation towards hosts, alighting, arrestment and feeding behaviour. To study the role of UV radiation on the flight activity of five insect species of agricultural importance (pests Myzus persicae, Bemisia tabaci and Tuta absoluta, and natural enemies Aphidius colemani and Sphaerophoria rueppellii), one-chamber tunnels were covered with six cladding materials with different light transmittance properties ranging from 2% to 83% UV and 54% to 85% photosynthetically active radiation (PAR). Inside each tunnel, insects were released from tubes placed in a platform suspended from the ceiling. Specific targets varying with insect species were placed at different distances from the platform. Evaluation parameters were designed for each insect and tested separately. The ability of insects to leave the platform was assessed, as well as the number of captures, eggs or mummies in each target, either sticky traps or plants. Our results suggest differences in flight activity among insect species and UV-blocking nets. The UV-opaque film drastically prevented aphids, and whiteflies from flying outside the tubes whereas T. absoluta, syrphids and parasitoids were not affected. Aphid flight behaviour was affected by the UV-opaque film compared to the other nets, especially in the furthest target of the tunnel. Fewer aphids reached distant traps under UV-absorbing nets, and significantly more aphids could fly to the end of tunnels covered with non-UV-blocking materials. Orientation of B. tabaci and T. absoluta was also negatively affected by the UV-opaque film although in a different trend. Unlike aphids, differences in B. tabaci captures were mainly found in the closest targets. UV transmittance did not have any effects on parasitoids, and S. rueppellii, implying cues other than visual for these insects under our experimental conditions. Further effects of photoselective enclosures on greenhouse pests and their natural enemies are discussed.
