  Abstract (down) Polyphenol oxidase (PPO) catalyses the oxidation of monophenols and/or o-diphenols to o-quinones with the concomitant reduction of oxygen to water which results in protein complexing and the formation of brown melanin pigments. The most frequently suggested role for PPO in plants has been in defence against herbivores and pathogens, based on the physical separation of the chloroplast-localized enzyme from the vacuole-localized substrates. The o-quinone-protein complexes, formed as a consequence of cell damage, may reduce the nutritional value of the tissue and thereby reduce predation but can also participate in the formation of structural barriers against invading pathogens. However, since a sufficient level of compartmentation-based regulation could be accomplished if PPO was targeted to the cytosol, the benefit derived by some plant species in having PPO present in the chloroplast lumen remains an intriguing question. So is there more to the chloroplastic location of PPO? An interaction between PPO activity and photosynthesis has been proposed on more than one occasion but, to date, evidence either for or against direct involvement has been equivocal, and the lack of identified chloroplastic substrates remains an issue. Similarly, PPO has been suggested to have both pro- and anti-oxidant functions. Nevertheless, several independent lines of evidence suggest that PPO responds to environmental conditions and could be involved in the response of plants to abiotic stress. This review highlights our current understanding of the in vivo functions of PPO and considers the potential opportunities it presents for exploitation to increase stress tolerance in food crops.

  Title Factors underlying changes in population of Phytophthora infestans in Poland Type

  Abstract (down) Phytophthora infestans (Mont.) de Bary belongs to Oomycetes and it causes the most destructive potato disease worldwide – late blight. It originates from Mexico but it has spread wherever potatoes are grown. P. infestans populations are diversified, sexual or asexual and their composition may be affected by climate changes. Mating type, mitochondrial haplotype, Simple Sequence Repeats (SSR) markers, sensitivity to metalaxyl and virulence were evaluated to monitor changes in Polish P. infestans population.Samples of potato leaflets with single late blight lesions were collected from fields located in three regions of Poland: Mlochów, Boguchwala and Siedlce, in three years 2010, 2011 and 2012. In the region of Mlochów intensively protected fields are dominating. There are mainly small gardens and experimental fields near Boguchwala. In Siedlce region early and starch potatoes are cultivated. Total number of isolates tested was 365. Mating type, mitochondrial haplotype and SSR were evaluated using a PCR method. Sensitivity to metalaxyl was tested on rye A agar media. Virulence was tested on detached potato leaflets.Polish P. infestans population is diverse. We did not observe major clonal lineages. A1 mating type (69%) and Ia mitochondrial haplotype (72.7%) dominated. Most of the isolates were sensitive to metalaxyl (66%). We noted differences in population composition between the regions which indicate that cultivation system has an impact on the population of P. infestans. No Label

  Abstract (down) Phytocystatins are a well-characterized class of naturally occurring protease inhibitors that function by preventing the catalysis of papain-like cysteine proteases. The action of cystatins in biotic stress resistance has been studied intensively, but relatively little is known about their functions in plant growth and defence responses to abiotic stresses, such as drought. Extreme weather events, such as drought and flooding, will have negative impacts on the yields of crop plants, particularly grain legumes. The concepts that changes in cellular protein content and composition are required for acclimation to different abiotic stresses, and that these adjustments are achieved through regulation of proteolysis, are widely accepted. However, the nature and regulation of the protein turnover machinery that underpins essential stress-induced cellular restructuring remain poorly characterized. Cysteine proteases are intrinsic to the genetic programmes that underpin plant development and senescence, but their functions in stress-induced senescence are not well defined. Transgenic plants including soybean that have been engineered to constitutively express phytocystatins show enhanced tolerance to a range of different abiotic stresses including drought, suggesting that manipulation of cysteine protease activities by altered phytocystatin expression in crop plants might be used to improve resilience and quality in the face of climate change.

  Title Multifractal analysis of meteorological time series to assess climate impact on chosen regions of Europe Type

  Abstract (down) Over the last decades modelling of climate change through the analysis of empirical meteorological data has become of great interest. The standard approach gives satisfactory results only in the climatic zones with extreme dynamics of climate change, thus there is need to develop and apply more subtle methods such as fractal analysis and chaotic evolution analysis of the atmospheric system. The scaling analysis of meteorological time series is complicated because of the presence of localized trends and nonstationarities. The objective of this study was to characterize scaling properties (i.e. statistical self-similarity) of the daily air temperature, wind velocity, relative air humidity, global radiation and precipitation through multifractal detrended fluctuation analysis on data from 31 years for stations located in Finland, Germany, Poland and Spain. The empirical singularity spectra indicated their multifractal structure. The richness of the studied multifractals was evaluated by the width of their spectrum, indicating considerable differences in dynamics and development. The log-log plots of the cumulative distributions of all the studied absolute and normalized meteorological parameters tended to linear functions for high values of the response, indicating that these distributions were consistent with the power law asymptotic behaviour. Additionally, we investigated the type of multifractality that underlies the q-dependence of the generalized Hurst exponent, by analysing the corresponding shuffled and surrogate time series. The results suggest that MFDFA is valuable for assessing the change of climate dynamics. No Label

  Abstract (down) Numerous climate futures are now available from global climate models. Translation of climate data such as precipitation and temperatures into ecologically meaningful outputs for managers and planners is the next frontier. We describe a model-based open platform to assess vulnerabilities of agricultural systems to climate change on pixel-wise data. The platform includes a simulation modeling engine and is suited to work with NetCDF format of input and output files. In a case study covering a region (Auvergne) in the Massif Central of France, the platform is configured to characterize climate (occurrence of arid conditions in historical and projected climate records), soils and human management, and is then used to assess the vulnerability to climate change of grassland productivity (downscaled to a fine scale). We demonstrate how using climate time series, and process-based simulations vulnerabilities can be defined at fine spatial scales relevant to farmers and land managers, and can be incorporated into management frameworks.
