Abstract: Phytophthora infestans is one of the most destructive potato pathogens. Many factors influence the population structure of P. infestans, including migration, climate and type of potato cultivation. Here, we analyse 365 P. infestans isolates collected from three regions of Poland over three years. We determined mating type, mitochondrial haplotype, resistance to metalaxyl, virulence and polymorphism at 14 simple sequence repeat (SSR) loci. Analysis of SSR markers showed high genetic diversity associated with this population. Model-based structure analysis grouped 299 unique genotypes into four main clusters. The P. infestans isolates collected from the Mlochow region, which has the most intensive level of potato cultivation, formed a distinct cluster, indicating a strong effect of the cultivation system on pathogen population structure. Three clusters contained isolates with frequent presence of three alleles at one locus, which may affect their capacity for sexual reproduction and preserve groups of fit genotypes that propagate asexually. Abstract: Phytophthora infestans (Mont.) de Bary belongs to Oomycetes and it causes the most destructive potato disease worldwide – late blight. It originates from Mexico but it has spread wherever potatoes are grown. P. infestans populations are diversified, sexual or asexual and their composition may be affected by climate changes. Mating type, mitochondrial haplotype, Simple Sequence Repeats (SSR) markers, sensitivity to metalaxyl and virulence were evaluated to monitor changes in Polish P. infestans population.Samples of potato leaflets with single late blight lesions were collected from fields located in three regions of Poland: Mlochów, Boguchwala and Siedlce, in three years 2010, 2011 and 2012. In the region of Mlochów intensively protected fields are dominating. There are mainly small gardens and experimental fields near Boguchwala. In Siedlce region early and starch potatoes are cultivated. Total number of isolates tested was 365. Mating type, mitochondrial haplotype and SSR were evaluated using a PCR method. Sensitivity to metalaxyl was tested on rye A agar media. Virulence was tested on detached potato leaflets.Polish P. infestans population is diverse. We did not observe major clonal lineages. A1 mating type (69%) and Ia mitochondrial haplotype (72.7%) dominated. Most of the isolates were sensitive to metalaxyl (66%). We noted differences in population composition between the regions which indicate that cultivation system has an impact on the population of P. infestans. No Label

Abstract: Phytocystatins are a well-characterized class of naturally occurring protease inhibitors that function by preventing the catalysis of papain-like cysteine proteases. The action of cystatins in biotic stress resistance has been studied intensively, but relatively little is known about their functions in plant growth and defence responses to abiotic stresses, such as drought. Extreme weather events, such as drought and flooding, will have negative impacts on the yields of crop plants, particularly grain legumes. The concepts that changes in cellular protein content and composition are required for acclimation to different abiotic stresses, and that these adjustments are achieved through regulation of proteolysis, are widely accepted. However, the nature and regulation of the protein turnover machinery that underpins essential stress-induced cellular restructuring remain poorly characterized. Cysteine proteases are intrinsic to the genetic programmes that underpin plant development and senescence, but their functions in stress-induced senescence are not well defined. Transgenic plants including soybean that have been engineered to constitutively express phytocystatins show enhanced tolerance to a range of different abiotic stresses including drought, suggesting that manipulation of cysteine protease activities by altered phytocystatin expression in crop plants might be used to improve resilience and quality in the face of climate change.

Abstract: Permanent grasslands provide a wide array of ecosystem services. Despite this, few studies have investigated grassland carbon (C) dynamics, and especially those related to the effects of land-use changes. This study aimed to determine whether the land-use change from permanent grassland to arable lands resulted in variations in the soil C stock, and whether such variations were due to increased soil respiration or to management practices. To address this, seasonal variations of soil respiration, sensitivity of soil respiration to soil temperature (Q(10)), and soil C stock variations generated by land-use changes were analyzed in a temperate mountain area of central Italy. The comparisons were performed for a permanent grassland and two adjacent fields, one cultivated with lentil and the other with emmer, during the 2015 crop year. Soil respiration and its heterotrophic component showed different spatial and temporal dynamics. Annual cumulative soil respiration rates were 6.05, 5.05 and 3.99 t C ha(-1) year(-1) for grassland, lentil and emmer, respectively. Both soil respiration and heterotrophic soil respiration were positively correlated with soil temperature at 10 cm depth. Derived Q(10) values were from 2.23 to 6.05 for soil respiration, and from 1.82 to 4.06 for heterotrophic respiration. Soil C stock at over 0.2 m in depth was 93.56, 48.74 and 46.80 t C ha(-1) for grassland, lentil and emmer, respectively. The land-use changes from permanent grassland to arable land lead to depletion in terms of the soil C stock due to water soil erosion. A more general evaluation appears necessary to determine the multiple effects of this land-use change at the landscape scale.

Abstract: Perception not only shapes knowledge but knowledge also shapes perception. Humans adapt to the natural world through a process of learning in which they interpret their sensory impressions in order to give meaning to their environment and act accordingly. In this research, we examined how farmers’ decision making is shaped in the context of changing climate. Using empirical data (face-to-face semi-structured interviews and questionnaires) on four Mediterranean farming systems from a case study located in Oristano (Sardinia, Italy) we sought to understand farmers’ perception of climate change and their behaviors in adjustment of farming practices. We found different perceptions among farmer groups were mainly associated with the different socio-cultural and institutional settings and perceived relationships between climate factors and impacts on each farming systems. The research findings on different perceptions among farmer groups can help to understand farmers’ current choices and attitudes of adaptation for supporting the development of appropriate adaptation strategies. In addition, the knowledge of socio-cultural and economic factors that lead to biases in climate perceptions can help to integrate climate communication into adaptation research for making sense of climate impacts and responses at farm level.
